The Effect of the Degree of Deacetylation of Chitosan on Blood Plumbum Levels (Pb), the Activity of the Aminolevulinic Acid Dehydratase (Alad) Delta Enzyme, and Hemoglobin Levels in Albino Mice (Mus Musculus L.)

Introduction

Environmental pollution by Plumbum (PB) is a serious problem caused by various human activities, such as mining, industries that use PB, and motor vehicle exhaust emissions. Pb exposure can cause various negative effects on health, including disorders of the nervous system, kidneys, and reproductive systems. One of the mechanisms of Pb's toxicity is inhibition of the activity of the Delta Aminolevulinic Acid dehydratase (Alad), enzymes that play an important role in the synthesis of hemoglobin. Therefore, it is essential to find a solution to mitigate the effects of Pb exposure.

Chitosan, a natural biopolymer from chitin deacetylation, has shown potential as a heavy metal antidote agent. The ability of chitosan in binding heavy metals is influenced by several factors, including the degree of deacetylation (DD). The higher DD, the stronger the ability of chitosan in binding heavy metals. This study aims to examine the effect of DD chitosan on blood PB levels, alad enzyme activity, and hemoglobin levels in mice exposed to PB.

Materials and Methods

This study involved 30 male mice strain balbc which was divided into 10 groups, with each group consisting of 3 mice. The control group consists of: K0 (Aquades Control), K1 (PB control), K2 (acetate control), and K3 (chitosan control). The treatment group consists of: P1 (Pb acetate + Chitosan A 1%), P2 (Pb acetate + Chitosan A 2%), P3 (Pb acetate + Chitosan B 1%), P4 (Pb acetate + Chitosan B 2%), P5 (Pb acetate + chitosan C 1%), and P6 (PB acetate + chitosan C 2%). The treatment is given orally using a Gavage needle with a volume of 0.01 ml/g BW for 14 days. The data obtained were analyzed using one-way ANOVA test and the BNT test (the smallest difference) to find out the significant differences between groups.

Results

The results showed that blood PB levels in the K1 group (0.33 µg/100 ml) were not significantly different (P> 0.05) with groups of P1, P2, P3, P4, P5, and P6. However, in general, the average blood PB level in the treatment group tends to decrease. This shows that chitosan with various degrees of deacetylation can reduce blood Pb levels in mice exposed to PB.

Alad enzyme activity in the K3 group (1.2 µmol Pbg/hour/l erythrocytes) increases significantly (p <0.05) compared to groups of K0, K1, K2, P1, P2, P3, P4, and P5. This shows that giving chitosan alone can increase the activity of the Alad enzyme. Alad enzyme activity in P6 groups is higher and significantly different (P <0.05) compared to groups of P1, P3, and P5. This finding shows that chitosan with high DD (P6 group) has a stronger effect in increasing the activity of the Alad enzyme.

Blood hemoglobin (HB) level in group K1 (12 g/dL) has decreased significantly (p <0.05) compared to all other treatment groups. This indicates that PB exposure can cause a decrease in Hb levels. Hb content in P6 groups has increased significantly compared to groups of P1 and P2. These results indicate that chitosan with high DD (P6 group) can help increase Hb levels in mice exposed to PB.

Discussion

This study shows that chitosan, especially with high DD, has the potential as a heavy metal bidder agent. Chitosan is able to reduce blood Pb levels, increase the activity of the Alad enzyme, and increase Hb levels in mice exposed to PB. The results of this study suggest that chitosan with high DD can be used as a potential antidote agent for Pb exposure. Further research needs to be done to examine the mechanism of work of chitosan in dealing with PB toxicity and identify optimal doses that are safe and effective for treatment.
